摘要
为探索痢疾杆菌感染是否产生免疫记忆,本文以福氏痢疾杆菌2a经口及腹腔免疫小鼠两次(相隔14d),间隔三个月后再刺激,应用BA-ELISPOT法检测了小鼠Peyer(PP)淋巴结、肠系膜淋巴结(MLN)及脾脏(SPL)中特异性IgA、IgG、IgM抗体分泌细胞(ASC)的变化;并以MTT比色法测定了ConA诱导的淋巴细胞增殖反应;还观察了小肠粘膜组织切片。实验表明:口服途径能诱导PP中ASC及淋转的回忆反应,小肠粘膜可见淋巴滤泡显著增生;而腹腔途径只在SPL中诱导出ASC及淋转回忆反应。结果提示:痢疾杆菌感染存在免疫记忆反应。
Mice were primed orally and intraperitoneally twice with an interval of 3months by shigella flexner 2a and boostered later on.The changes of the antibody secreting cells(ASC)for specific IgA.IgG.IgM in Peyer's patches,mesenteric lymphonodes and spleen of mice were determined by BA-ELISPOT method,and the proliferation responses of lymphocytes induced by Con-A were assayed by MTT colorimetic method.It was found that the memory responses of lymphocyte proliferation were induced by oral route with a significant lymphofollicle proliferation in the intestinal mucosa,but the induction of ASC and the memory responses by intraperitoneal toute were only found in the spleen,suggesting the existence of memory responses during infection with dysentery bacilli.
