Effeet of the Cytoskeletal System on TNF-α Production

摘要 本文证实细胞微管聚会抑制剂秋水仙碱、长春花碱、长春新碱等均可明显抑制脂多糖刺激的大鼠腹腔巨噬细胞合成肿瘤坏死因子-α(TNF-α)。微管解聚抑制剂重水与Taxol亦可使TNF-α产生轻度减少,提示微管的聚合与解聚对TNF-α的正常产生都非常重要,其中聚合功能对TNF-α合成的调节尤其重要。此外,微丝聚合抑制剂细胞松弛素B亦可轻度抑制TNF-α的产生,说明微丝亦参与对TNF-α合成的调节。 Microtubule polymerization-disruption agents such as colchicine, vinblastine,vincristine and nocodazole have been shown to markedly inhibit LPS-induced TNF-αproduction by rat peritoneal macrophages(Mφ).Microtubule depolymerization-inhibiting agents deterium oxide and Taxol were also found to reduce TNF-αproduction,albeit to a much lesser extent. These observations suggest that both polymerizationand depolymerization of microtubules are necessary for normal TNF-αproductionwhile the former seems to play a more important role in the regulation of TNF-αsynthesis. Besides, cytochalasin B, an inhibitor of microfilament polymerization,showed a slight suppression of TNF-αproduction as well, denoting that microfilaments are also involved in the regulation of TNF-αproduction by the Mφ.
