摘要
稳定分泌抗IBDV单克隆抗体的杂交瘤细胞(I—4—3),培养于5LCelligen中,接种浓度不少于2x109/L,杂交瘤细胞在Celligen中生长,随着细胞数浓度的变化,其生物学和动力学参数发生相应的变化,杂交瘤细胞在PH7.18、DO50、50~70r/min及葡萄糖浓度为2.5g/L下生长良好。每天可收集3L的单抗,结合抗体定量试验,测得具有生物学特性的抗体含量为300mg/L以上。研究结果表明5LCelligen可用于杂交瘤细胞在体外大规模生产单克隆抗体。
Hybridoma cell (I-3-4) which could Stable secrete monclonal antibody to IBDV was cultured in SL celligen where the cell density was more than 2x109/L. The kinetic and biological parameters also changed with the change of the cell density. The hybridoma cell grew well at PH 7. 18,DO 50, 50 ̄70r/min when the concentration of glucose in culture medium is 2. 5g/L, 3L/day McAb with its concentration of 300mg/L,could be collected. It was concluded that hybridoma cell in 5L celligen could successfully produce McAb in vitro in large scale.
