摘要
用瞬时甲基化气液色谱法(GLC)分离和测定脑组织中存在游离脂肪酸(FFA,包括棕榈酸C16:0、油酸C18:1、硬脂酸C18:0、花生四烯酸C20:4、廿二碳六烯酸C22:6)。最低检测限为0.028~0.045nmol,用十五烷酸C15:0作内标,测得5种标准脂肪酸的线性范围为0.11~1.74nmol,相关系数r=0.9992~0.9999,方法回收率为95.36%~98.77%,天内精密度RSD<5.60%;天间精密度RSD<5.79%。
A convenient and reliable flash methylative gas-liquid chromatographic methed for determining free fatty acids(FFA)in brain tissue specimens was studied.The limit of detection of FFA was 0.028~0.045 nmol.With pentadecanoic acid(C15:0)as an internal standard,a linear relationship was from 0.11~1.74 nmol(five FFA,r=0.999 2~0.9999).The overall percentage recovery of FFA was 95.36%~98.77%.Their RSD were<5.60% within day,and<5.79% between days.
