MTT法分析培养成骨细胞的存活和增殖能力 被引量:79

USE OF MTT ASSAY FOR DETERMINATION OF OSTEOBLASTS VIABILITY AND PROLIFERATION IN VITRO

摘要 用MTT法测定离体培养的成骨细胞存活与增殖能力。并观察137Csγ线对成骨细胞生长的影响。细胞接受0、3、6、9、12Gy照射,培养至第7天,增殖率分别为416、336、275、147、108%。结果表明:137CSγ线剂量高于3Gy成骨细胞增殖受到显著抑制,9Gy以上,则基本抑制其增殖。 Taking advantage of rapid and simple colorimetrie cell viability assay, we employed MTT assay observing the growth of osteoblasts.Conditions for determination were(1)serum-free medium was used to avoid interruption in tes; (2) samples were:measured at wave length 570nm;(3) four hours was required to form formazan; and(4) the linearity was obtained at range of cell number 30 × 103 ̄1×106(r=0.9701). In addition, the result of MTT assay was consistent with that of cell counting.(Radiation can significantly suppress growth of osteoblasts.The proliferation rates were 416% ,336%,275%,147%,108% at groups of cells exposed to 0,3,6,9, 12Gy respectively after seven-day culture. Results suggest that the cell growth can be significantly suppressed with the dosage above 3 Gy from 137Cs radiation and be completely blocked after more than 9 Gy radiation.
