摘要
本试验用0.1mL培养液在37℃,5%CO2条件下培养昆明小鼠原核胚。结果表明,用mSOF和PL3培养小鼠原核胚,4-细胞发育率分别为10%和77%,以后停止发育;用PL3和PL3’培养小鼠原核胚,每24h换1次培养液,结果发育到桑椹胚和囊胚的百分率分别是27.5%和38.5%;用mMTF,mMTF+20AA,mMTF+Non+Gln,mMTF+Non,mMTF+Ess,mMTF+Ess+Gln培养小鼠原核胚,发育率分别为57.1%,51.0%,56.7%,58.1%,59.1%和54.9%,各组间差异不显著(P>0.05)。试验证明,几种培养液均可使小鼠原核胚克服阻断,继续发育,但改换培养液有利于其进一步发育;氨基酸对小鼠胚胎发育有促进作用.
The result obtained from chemical media at 37℃ in 5% CO2 in air on in vitro development of Kunming mouse pronuclear embryos were that the developmental rates were 15% and 77%, resp., with significant difference (P>0.001)during cultured with mSOF and PL3; the developmental rates were 27.5% and 38.5%,resp. when cultured in PL3 and PL3 and replacing with fresh media one time per 24h;the mouse pronuclear embryos cultured in mMTF+20 AA, mMTF+Non+Gln,mMTF+Non, mMTF+Ess and mMTF+Ess+Gln got the developmental rates of 57.1%, 51.0%,56.7%, 58.1%, 59.1% and 54.9%, resp., with no significant difference (P>0.05). The above results demonstrate that chemical media can support in the development of mousepronuclear embryos.Amino acids can play a role on mouse embryonic development.
