摘要
利用克隆系NG108-15细胞在膜片箝全细胞(whole-cellclamprecording)记录下,观察了国产TTX单克隆抗体8A5拮抗TTX对跨膜钠电流的作用。结果表明,8A5有效地抑制TTX的作用:(1)在标准灌流液中加入1μmol/LTTX,Na+流立即被完全阻断,复加1μmol/L8A5于灌流液,可使Na+流迅速恢复至对照水平;(2)若先以含1μmol/L8A5的溶液灌流细胞,再更置为含1μmol/LTTX的标准液,Na+流只有部分被抑制。
Anti-TTX effect of 8A5 on Na+ current was observed by whole-cell voltageclamp technique in NG108-15 cells. The results show that 8A5 inhibits TTX action efficiently: (1) When 1 μmol/L TTX is added to the normal perfusion solution ⅠNa disappears immediately, and can be restored to the control level with further addition of8A5. (2) When the cell is pre-incubated with 1 μmol/L 8A5, 1 μmol/L TTX can only cause a partial inhibition of ⅠNa.
