摘要
用分子杂交技术及流式细胞方法研究了转化生长因子α(TGFα)在人胃肠道肿瘤组织中的表达.结果表明,胃癌、结肠癌和直肠癌组织较相应癌旁组织TGFαmRNA水平增高.佛波脂(TPA100μg/L)作用胃癌细胞MGc80-3后4h,可使细胞中TGFαmRNA水平增高,作用6h可使S期细胞百分比明显增加.这些结果提示:TGFα在胃肠道肿瘤中起重要作用,其表达可受TPA的调节.
The expression of transforminggrowth factor α(TGFα)in human gastroin-testinal carcinoma by methods of molecularhybridization and cell FIT cell-cycle analysiswas investigated.The results show that thelevels of TGFα mRNA are higher in gastriccarcinoma tissues,colonic carcinoma tissuesand rectal carcinoma tissues than in the corre-sponding control tissues. After treating with12-O-tetradecanoylphorbol-3-acetate(TPA,100μg/L,4h),the level of TGFαmRNA ingastric carcinoma cell line(MGc80-3)increas-es, and percentage of S phase cells is marked-ly increased after 6 hours.These results sug-gest that TGF α plays an important role ingastrointestinal carcinoma tissues and expres-sion of TGFα mRNA can be regulated byTPA.
