多药抗药基因Mdrl探针的克隆及初步应用 被引量:2

摘要 多药抗药基因Mdrl的表达水平与细胞的耐药性直接相关,检测Mdrl的表达水平可预测化疗的效果以及预后,用分子原位杂交的方法可检测单个细胞中Mdrl的表达水平.用PCR扩增方法获得了一段特异的DNA片段,并将其克隆到pUC18载体中,经DNA序列分析证明与文献报道一致,此探针可用于临床标本的分子杂交检测. Mdrl gene contributes to multidrugresistance.Because of the level of Mdrl geneexpression is correlated with the response ofchemotherapy,it is a valuable method to mea-sure the expression level of individual pa-tients. The cDNA cloning of the human Mdrlgene has made it possible to measure levels ofMdrl RNA with hybridization in human can-cer cells. A special part of Mdrl cDNA wasobtained by use of RT-pCR and inserting itinto pUC18 vector.The DNA sequencingresult is identical with the sequence of MdrlcDNA reported.This probe can be used to de-tect the level of Mdrl gene expression in clini-cal specimen,The measurements can be usefulin the design of chemotherapeutic protocolsfor certain tumors.
