摘要
用定点突变法分别得到了两个人白细胞介素-2(IL-2)的部分拮抗剂15Val-IL-2和126Asp-IL-2以及一个为IL-2受体α亚基结合缺陷型的突变体62Leu-IL-2,当将15Val-IL-2或126Asp-IL-2与62Leu-IL-2共同保温时,62Leu-IL-2的活性受到明显抑制,对此现象机理的分析表明15Val-IL-2或126Asp-IL-2可用于IL-2受体亚基结合缺陷型突变体的初步鉴定.同时,这一思路在其它受体-配基系统中具有一定的适用性.
Two partial antagonists of humaninterleukin-2(IL-2):15 Val-IL-2,126Asp-IL-2 and an IL-2 receptor a subunit binding defi-cient mutant 62Leu-IL-2have been obtainedby site-directed mutagenesis, 62Leu-IL-2could exhibit significant stimulation on CTLL-2cells. When 15Val-IL-2 or l26Asp-IL-2 wasincubated together with CTLL-2 cells in addi-tion to 62Leu-IL-2,the stimulative ability of62Leu-IL-2 was distinctly inhibited.In con-sideration with the interaction of IL-2 with IL-2 receptor subunits,15Val-IL-2 or l26Asp-IL-2 can be used for the primary determina-tion on receptor subunit binding ability of IL-2mutants,Furthermore,this idea may also beapplied in some other ligand-receptor sys-tems.
