外源性GM_3/GD_3对人肝癌培养细胞原癌基因表达的研究

摘要 应用原位分子杂交技术及细胞图象分析证实了人肝癌细胞株SMMC7721细胞经10μg/ml的GM_3/GD_3处理后,GD_3处理组细胞内N-ras基因mRNA的阳性信号弥漫分布于整个细胞,而经GM_3处理组和对照组阳性信号集中于核膜。GM_3/GD_3处理组及对照组c-fos基因mRNA的阳性信号均集中于核膜,但GM_3处理组信号强度最大。上述结果提示,GD_3可使某些肿瘤细胞向更加恶性的方向发展,而GM_3则可使某些肿瘤细胞向正常方向转化。 With the method of in situ hyridyzation and cell image analysis, it was showed that the positive signal of N-ras mRNA in the GD_3-treatcd cells distributed broadly within the whole cell, whereas that of GM_3-treated and control cells were concentrated on the nucleus membranes. The positive signals of c-fos mRNA in nucleus membranes, but GM_3-treated cells has the strongest one. The results mentioned above indicated that GD_3 could induce some tumer cells to be more malignant, but GM_3 could reverse some of the effect.
