摘要
研究了人工合成的谷胱甘肽过氧化物酶(GSHPx)模拟物-ebselen及其辅助因子谷胱甘肽(GSH)对Cu2+及紫外线(UV)诱导的低密度脂蛋白(LDL)氧化修饰的抑制和终止作用。结果显示:在GSH存在的情况下,低主5-μmol/L的ebselen就可完全抑制由5μmol/LCu2+引发的LDL氧化修饰,单独的ebselen和GSH没有抑制作用,50μmol/L的维生素E只有很微弱的抑制作用。Ebselen对Cu2+引发的LDL氧化修饰的抑制作用时间的长短与GSH的浓度有关。对已由Cu2+引发启动的LDL氧化修饰,加入ebselen和GSH可防止其进一步的氧化。Ebselen和GSH对紫外线引起的LDL氧化修饰也有一定的抑制作用。
Inhibition and termination effect of a synthesized glutathione perendase rnnmc-ebselenand its cofactor GSH on the oxidative moderation of lOw density lipoproch (LDL) studied by detemnination of thiobarbituric and reactive substance gnase) and fluorescence intensity. The results showed that ebselen at a low concentration of 5μmol/L could inhibitoxidative modistion of LDL induce by saal/L Cu2+ completely in the presence of GSH(50μmol/L). Neither ebselen nor GSH alone had the effat, Vitamin E (50μmol/L) had only avery weak effed. The the of inhibition of ebeelen on the oxidative modistion of LDL induced by Cu2+ was dependent on the conCentlation of GSH. Ebselen together with GSH couldprevent further oxalation of LDL. As to the oxidahve rnodfotion of LDL induce by UVinadtation, to some extent an inhibitory effat of ebeelen and GSH was shown.
