摘要 用无乳化剂聚合法制备了苯乙烯-甲基丙烯酸-β-羟丙酯-丙烯酸(st-HPMA-AA)三元共聚胶乳。讨论了其聚合条件与影响因素。通过硝化反应与还原反应在苯乙烯的苯环上接上氨基,利用重氮化反应将登革热抗体与之相交联制成登革病毒胶乳凝集免疫诊断试剂。试验表明该诊断试剂对登革病毒以外的其它虫媒病毒无明显交叉反应,最低检出抗原量达到15ng/ml病毒蛋白水平,敏感性是血凝方法的100倍。经临床诊断表明,该试剂具有特异性好,诊断快速、简便及敏感性强等优点,为登革热的早期快速诊断和监测提供了一种有效方法。 Ternary copelymer latex St-HPMA-AA was synthesized by means of emulsion-free polymer reaction method. The effects of various parameters on polymer reaction were discussed. Ternary copolymer ammoniacal latex was obtained using the reaction of nitration and reduction reaction. With the diazonium reaction. the antibody against Dengue virus was conjugated with this ammoniacal latex, and a rapid diagnosis immunological reagent against Dengue virus was obtained. The experiments showed that the reagent has little cross-reaction with other arbovirus. The sensitivity of diagnosis is about 15ng/ml, 100 times higher than that of hemagglutination assay.Clinical diagnosis confirmed that this immunological reagent is rapid, convenient and sensitive on diagnosis of Dengue virus.
