摘要
本实验将载有AR-cDNA的重组质粒转化入感受态JM109宿主菌内,通过扩增、酶切、纯化,成功地制备了540bp的AR-cDNA片段,应用“随机引物”法制备AR-cDNA探针,经Northerl。blot分析,结果可见,睾丸组织有雄激素受体mRNA的表达。本研究为探讨某些生理病理条件下睾丸AR基因水平的变化奠定了基础。
In the present experiment, the recombinant plasmids with AR-cDNA (androgen recepter cDNA)were transfered into competent JM 109 host bacteria. With the recombinant plasmids having been amplified, digested with restrict endonueclase and purified, 540bp cDNA fragment were recollected successfully. The AR-cDNA fragment were labelled with α-32P prime-α- gene. The results showed that there was an expression of AR- mRNA in testicular tissue. This results are of significant importance in exploring changes of testicular AR gene level under physiological and pathologic conditions.
