Effect of HSG2 Expression Correspondent Antibody on Sperm's Motility

在构建成人睾丸ｃＤＮＡ文库的基础上，用兔抗人精子抗体自文库中筛选出了８株阳性克隆，并对其中的ＨＳＧ２表达产物进行理化特性测定和抗ＨＳＧ２表达产物抗体的生物学活性测定。ＳＤＳ－ＰＡＧＥ电泳显示溶原蛋白的分子量约为１５５ＫＤａ。抗ＨＳＧ２表达产物抗体能明显的抑制精子的运动能力，对精子凝聚、顶体蛋白酶和精子表面的ＣｏｎＡ受体无影响。提示ＨＳＧ２表达产物可能成为精子疫苗的候选成份。

After having isolated 8 human sperm gene clones(HSG1-8) with rabbit'S anti-sperm antibodies as probes, we determined physical and chemical nature of HSG2 expression and anti-fertility effects of expression correspondent antibody.SDS-PAGE electrophoreses showed thatmolecular weight of lysogen was 155 Da.Expression correspondent antibody could inhibit themotility of human sperm. But there were no effects on sperm's agglutination,acrosin activity andCon-A receptors on sperm's surface.The results indicated that HSG2 expression could be usedas candidate for sperm vaccine and as coating antigen in ELISA for the detection of anti-spermantibody.
