摘要
本文用ＰＥＧ６０００沉淀１０ｍｌ健康人混合精浆，粗提ＰＳＡ，再通过二次ＳｅｐｈａｏｌｅｘＧ－１５０柱层析，获得纯化的ＰＳＡ．经ＳＤＳ－ＰＡＧＥ分析为分子量约３４０００ｄｏｌｔｏｎ的单一条带。ＥＬＩＳＡ法测得含量为１．１０ｍｇ／ｍｌ，考马斯亮蓝法测其蛋白浓度为１．１４ｍｇ／ｍｌ．本法操作简便，所得产物经免抗人精浆血清证实达到免疫纯。
A crude extroction of prestate specfic antigen (SPA) was precipitated by PEG6000 from 10ml mixed seminal plasma of healthy adults,then pure PSA was obtained by twice sephadex G-150 column chromatography. SDS-PAGE analysis showed a single band of molecular weight about 34000 dalton. Concentration of PSA and protein in end product were 1.10 mg/ml and 1.14 mg/ml using ELISA and Coomassie blue test, respectively. This method was simple,The finished product reached immunological purity confirming by immunoelectrophoresis with rabbit anti-human sperm plasma serum.
