高压液相色谱法测定阿米替林及其代谢产物的浓度 被引量:3

摘要 应用高压液相色谱法测定人体血浆内阿米替林及其主要代谢产物去甲替林及羟基去甲替林的血药浓度。以产国硅胶YQG—80(5μm)为固定相,甲醇—水极性流动相,三甲丙咪嗪及普罗替林为双内标物。紫外检测波长240nm。阿米替林、去甲替林和羟基去甲替林的最低检出限量分别为2,5和5ng,最低检出浓度分别为4,10和10ng/ml(血浆),回收率分别为>94,97和97%。方法简便,灵敏度高,适于临床药理研究及临床治疗监测。 A high-performance liquid chromatomato method was deve-loped to determine plasma levels of amitriptyline and its two major meta-bolites, nortriptyline and 10-hydroxynortriptyline. Silica gel (YQG-805μm, produced in Qingdao, China) was used as solid phase and methanoland water as mobile phase. Trimipramine and protriptiline were used astwo internal standards. The wave-length of UV detector was 240 nm. Thelowest detection limits of amitriptyline, nortriptyline and 10-hydroxynor-triptyline were 2, 5, and 5 ng, respectively, and that of plasma concen-trations were 4, 10, and 10ng/ml, repectively. The recovery rates were>94, >97, and >97%, respectively. For its convenience and sensitivity,the method is suitable for clinical pharmacological research and conven-tional therapeutic monitoring.
