摘要
本文采用中华大蟾蜍卵母细胞(BBO)体外翻译系统研究了不同来源细胞IL—2RαmRNA的翻译活性。显微注射mRNA的BBO用Barths液经20℃培养可获得最高翻译效率。与非州爪蟾卵母细胞的比较表明,两系统的体外翻译效率相近。对PHA刺激的人、脾淋巴细胞IL—2RαmRNA的合成动力学的观察表明,IL—2RαmRNA刺激后6小时即出现,9小时高达高峰,随后逐渐下降。放射受体分析显示,注射IL—2RαmRNA后,IL—2Rα也可表达于BBO膜表面。
The translation activities of IL-2Ra mRNA among different kinds of cells by using Bufo bufo oocytes (BBO) in vitro were studied. The highest translation efficiency was observed when BBO were cultured in Barth's buffer at 25℃. The translatbn activities of IL-2 R a mRN A in both BBO and Xenopus laevies oocytes (XLO) systems were found to be nearly the same. The analysis on synthetic kinetics of IL-2Ra mRNA of human spleen lymphocytes showed that IL-2 R a mRNA appeared 6hr(?) after PHA stimulation and reached its peak 9 hrs after and then gradually declined. The radioreceptor assay showed that the IL-2Ra can be also expressed in oocyte membrane after microinjection of IL-2Ra mRNA.
