摘要
用基因重组人IL—2(rIL—2)免疫的BALB/C小鼠脾细胞与NS—1细胞株杂交,经3~5次克隆获得9株分泌抗IL—2单克隆抗体杂交瘤,所分泌抗体均为IgG_1。其中3B5杂交瘤诱生的腹水中抗体效价达10万倍稀释度。它既能中和用于免疫的rIL—2活性,也可中和其它来源的基因重组人rIL—2,但与天然rIL—2反应弱。rIL—2可与4C3抗体包被的Sepharose 4B组合,並能用1.5%醋酸洗脱。因此它可做为基因工程制备rIL—2的辅助试剂及用于与rIL—2相关的研究。
Spleen cells of BALB/c mice immunized with recombinant human IL-2 (rIL-2) were hybridized with NS-1 myeloma cell 1ine. 9 stable clones of hybri domas secreting monoclonal antibody (McAb) against human IL-2 were obtained. The isotype of the McAb was defined as (?). The antibody titer in 3B5 clone-induced ascite was found to be higher than 10^(-5). This McAb was able to neutralize the activity of the immunizing rIL-2 (Cetus) as well as human rIL-2 (expressed in E. coli and yest) from other sources. It was found that this McAb reacted weakly with human natural IL-2 and the recombinant IL-2 avidly bond to an affinity column of Sepnarose 4B conjugated with the McAb and could be eluted with 1.5% acetic acid. Thus these McAbs are useful for the preparation of purified rIL-2.
