摘要
本文以轮状病毒(RV)Wa株和SA-11株为代表,采用国产16孔组织培养板,进行了RV半微量空斑技术的研究,并与常规培养瓶空斑法和TCID_(50)滴定法作了比较。结果表明,RVWa株与SA-11株在MA_(104)细胞上的空斑特征和感染滴度有较大差异,半微量法空斑数目与病毒浓度呈良好的线性关系,且重复试验能获得相近空斑滴度,显示了经济方便、稳定可靠的优点。此外,应用空斑中和试验测定了RV型特异性抗血清的中和效价,并首次注意到用活性染料中性红染色的空斑标本经福尔马林固定后可长期保存不褪色。
A semimicro plaque technique of rotavirus ( RV ) was developed with the model of RV Wa strain and SA-11 strain in 16-well culture plates and compared with the routine plaque technique in culture bottles and TCID50 method There were distinct differences between Wa strain and SA-11 strain in plaque morphology and infectious titer. The linear relationship between pla que number and virus concentration was obtained by the semimicro method. The similar titers of a preserved virus stock were given by the repeated assay cells. All these results suggested that the semimicro plaque technique should be of great economy, convenience and reliability. Furthermore, the type -specific neutralizing antisera to Wa strain and SA-11 strain were titrateb by the semimicro plaque neutralization test.It was first found that the plaque samples with neutral red staining fixed with formalin could be preser ved for a long time.
