在体外培养系统中中性粒细胞对日本血吸虫童虫的作用

摘要 3小时童虫在含病兔血清、补体和中性粒细胞的培养系统(ISCN)中培养48小时后,约60%的童虫体表有中性粒细胞附着,其中50.8%的童虫已死亡,含病兔血清不含补体(ISCN)、含正常小鼠血清及补体(NSCN)或含正常血清不含补体(NSN)的培养系统中,中性粒细胞粘附童虫体表较少,童虫的死亡率亦低。24小时童虫在ISCN中培养48小时后,中性粒细胞对童虫的附着及童虫死亡率均逊于3小时童虫。在ISCN中,中性粒细胞对72小时童虫几不附着。以中性粒细胞介导的杀死3小时童虫的作用依赖于抗体的存在,但不完全依赖于补体。 3-h old schistosomuia of Schistosoma japonicum were maintained in four culture systems for 48h. When the culture system containing infected rabbit serum, complement and neutrophils (ISCN) was used,neutrophils adhered to about 60% of schistosomuia, while the mortality of the worm was 50. 8% . In other culture systems containing neutrophils and infected rabbit serum without complement (ISN) , neutrophils and normal mouse serum with complement (NSCN) or neutrophils and normal serum without complement (NSN) ,less schistosomuia were adherent with neutrophils on their surface and lower or less mortality of the worm were observed. When 24-h old schistosomuia were maintained in ISCN for 48h, the percentages of the worm, attached and killed by neutrophils were less than those seen in 3 - h old schistosomuia maintained in corresponding culture system. When 72-h old schistosomuia were maintainde in ISCN for 48h there were not any attachment of neutrophils on most of the worm . The results suggest that the neutrophil-mediated cytoxicity on 3 -h old schistosomuia should be dependent on specific antibody and partially dependent on complement.
