单克隆抗体铺皿法分离人外周血淋巴细胞亚群

Separation of Lymphocyte Subsets by Using McAbs

应用2种单克隆抗体(McAb CD_1^+、CD_3^+),通过 Panning 直接法和间接法分别分离人外周血相应的淋巴细胞亚群,间接免疫荧光试验和台盼蓝拒染分别鉴定细胞的纯度和存活率,对两种方法所分离的纯度和产额、存活和丝裂原刺激反应作了比较。结果表明直接法分离的亚群细胞的纯度略低于间接法。

McAb CD_4 ,CD_8 were used to separate CD_4,CD_8 cells from human peripheral blood mononuclear cells by direct or indirect'Panning' methods.The purities,yields,viabilities and reactions to mitogen -PHA of CD_4,CD_8 cells separated by two methods were compared.The results indicated that the purity of CD_4,CD_8 obtained by direct method was lower than that by indirect method.But in respect to the viabilities,yields and reactions to PHA,direct method was better than indirect method.An inverse relationship was found between the yield and purity of subsets.
