摘要
应用LC一6A型高效液相色谱仪,建立了快速测定唾液中甲硝唑及其代谢产物的方法。固定相为Shim-pack CLC-ODS柱,甲醇-水(27:73)作流动相,流速1.2ml/min,检测波长3l8nm。甲硝唑平均回收率为99.40%,日内和日间变异系数分别为3.72%和3.98%。最低检出浓度为0.025μg/ml,线性范围0.05~20μg/ml(r=0.9999)。对唾液与血清中甲硝唑浓度的相关性作了初步探讨。
A reversed一phase HPLC method has been developed for rapid determination ofmetronidazole and its metabolite in human saliva by using a Model LC一6A HPLC with two pumps anda Shim一pack CLC一ODS colum. The mobile phase,mixture of methanol and redistilled water(27 :73,byvol),was used at a flow rate of l.2ml/min. The samples was monitored at 3l 8 nm wavelength. Themean recoverv of metronidazole in saliva was 99.40%,Within一day and between一dav variations were3.72%and 3.98%,respectively. The measurable lowest level was 0.025 μg/ml and the standard curve ofmetronidazole was linear over the concentration range of 0.05~20 μg/ml(r=0.9999 ).The pharma-coknetics of metronidazole in human after oral administration and the relativity between saliva andserum concentrations were studied.
