摘要
本文对Renz等[1]直接酶标HBVDNA探针检测乙肝患者血清HBVDNA法进行了改进,用其直接酶标HBVDNA制备探针的方法,杂交后用增强化学发光检测系统取代直接酶标DNA探针检测法中的酶催化底物显色法[2]。结果显示,改进后的方法在保持了特异性的同时,大大地提高了直接酶标HBVDNA探针检测法的敏感性,使这种方法成为可替代同位素探针测定血清中HBVDNA的一种新的快速简便的检测方法。
In the article,Renz's methods for detection of HBV DNAin serum were modified. The dot blot hybridization was carried out using Renz's direct enzyme labelled HBV DNA probes.After hybridization the HBV DNA was visualized by enhanced chemiluminescence system that replaced an enzyme-linked color reaction performed in the dark.The modified method raised significantly the sensitivity and specificity of the Renz's method.The method is available alternative to the radiolabeled probe for the detection of HBV DNA in serum.It is a new,fast,simple and convenientmethod.
