刺五加多糖及谷胱甘肽硫转移酶对大鼠实验性肝癌防治的研究 被引量:7

Study on Role of Acanthopanax Senticosus Polysaccharides and Glutathione S-transferase during 3'MeDAB Hepatocarcinogenesis

摘要 应用甲氨基偶氮苯(3'-MeDAB)饲喂大鼠造成实验性肝癌模型,同时饲喂刺五加多糖口服液(剌五加组)和用自制的电泳纯人胎盘谷胱甘肽硫转移酶(GST组)免疫大鼠,历时110天。结果:1.各实验组大鼠肝脏病理组织学改变以非典型增生为主,癌形成率平均为6.7%,以GST组病变最重,癌形成率为17%,经统计分析x^2=16.81,P<0.01。免疫组化检测肝脏γ-谷氨酰转肽酶(γ-GT)阳性颗粒的出现率及程度(++)与上述病理改变吻合,经统计学分析x^2=12.03,P<0.05。2.γ-GT活性:饲喂3'-MeDAB 4天即有明显升高(2.3倍),实验结束时平均可达对照组的40倍。GST组可高达80~90倍。3.肝脏GST活性及GSH含量:各实验组较对照组均明显减少,GST组最显著。方差分析F=4.96,P<0.01,q检验3'-MeDAB组与对照组相比,q=2.87,P>0.05,而GST组和刺五加组分别与对照组相比,q=4.74和q=4.65,P<0.05和P<0.01,各实验组之间相比,均P>0.05。未观察到刺五加预防实验性肝癌形成的效果。 In normal rats administration of diet containing 3'-MeDAB for more than 10 weeks resulted in development of liver tumors. In order to study the role of acanthopanax senticosus polysaccharides and glutathione S-transtferase (GST) during 3'-MeDAB hepatocarcinogenesis ,rats were immunized with GST of human placenta in one group (GST group).and fed with water containing 20% acanthopanax sentirosus polysaccharides to another group. During the study period diet containing 3'-MeDAB were given simultaneously to both groups. (1) γ-GT immunohistochemical localization was detected (ABC method).γ-GT postivity of liver was 78% in the GST group and main hepatic changes were focal nodular hyperplasia.with only 17% developing hepatomas. (2) The level of γ-glutamyl transpeptidase (γ-GT) of rat liver increased significantly after the 3'-MeDAB diet for 4 and 110 days,it showed an extremely high activity approximately 40 times higher than the controls .especially the GST group which showed 80-90 times as high as the controls.lt is now considered to be one of the best markers for the preneoplastic lesion of experimental hepatoma and employed as a tumor marker for the hepatoma. (3) GST levels and total glutathione content in the liver of all experimental groups were lower than that of the controls.
