摘要
本实验观察了猪离体冠状动脉(CA)的缺氧反应。缺氧可使CA收缩,血管张力提高1.56±0.43g(n=13).在以组胺(2×10-6mol/L)使CA具有一定主动张力后,缺氧使血管张力提高3.01±0.68g,显著高于用组胺前之反应(P<0.01)。去除血管内皮对CA的缺氧性收缩无影响(n=13,P<0.05)。维拉帕米显著抑制了CA的缺氧性收缩反应;吲哚美辛、乙胺嗪、过氧化氢酶、SOD、U-63557A(TXA2合成抑制剂)无作用,结果揭示缺氧可引起离体冠状动脉的收缩,这一反应不依赖于血管内皮,而是与缺氧直接作用于血管平滑肌、促使钙内流或细胞内钙的释放所致。
The hypoxic response of porcine coronary artery rings(CA) with or without endothelium was studied in quiescent state or by precontracting with histamine(2x10-5 mol/L).Hypoxia induced vasoconstriction in both quiescent CA and precontracted ones,the tension increased by 1.56±0.43g(n=13) and 3.01±0.68g(n=13) respectively with the latter being significantly higher than the former(P<0.01).The hypoxia vasoconstricition remained unchanged in either quiescent or precontracted end otheliumdenuded artery rings (n=13, P>0.05).Calcium antagonist verapamil markedly inhibited the contraction caused by hypoxia in CA with or without endothelium,whereas indomethacin,diethylcarbamazine,catalase and superoxide dismutase,U-63557A(a TXA2 synthesis inhibitor) did not show such effects.The results indicate that,the hypoxic constriction of CA is endothelium-independent and related to the direct effect of hypoxia on the Ca2+ influx of smooth muscle cells.
