摘要
采用^3H-TdR、^3H一脯胺酸、DNA定量检测及MTT比色等方法,研究猪肺动脉缺氧性内皮细胞条件培养液对猪肺动脉平滑肌细胞增生及胶原蛋白合成的影响。并探讨丹参对此过程的阻抑作用。结果表明:HECCM能引起PASMC的线粒体酶活性升高,核内DNA含量增加及^3H-TdR、^3H一脯氨酸掺入量增多。
The effects of hypoxic endothelial cell conditioned Medium(HECCM) onproliferation and collagen synthesis of cultured porcine pulmonary smooth muscle cell (PASMC) were studied by 3H-thymidine and 3H-proline incorporation,image analysis for determination of DNA content and colorimetric assay using MTT,and the inhibitory effects of Radix salviae miltiorrhizae(RSM) on them were also investigated. The results showed that HECCM could induce enhancement of the enzymatic activity of mitochondria, increase of the nucleic DNA content and increase of the  ̄3H-TdR and  ̄3H-proline incorporation in PASMC.The 3H-proline incorporation in PASMC cultured in HECCM was 1.83 times as much as that cultured in NECCM, Compared with the control chinese herb medicine RSM could inhibit proliferation of PASMC cultured in HECCM and decrease the  ̄3H-proline incorporation in PASMC cultured in both HECCM and NECCM(P<0.01),respectively. However, RSM had no effects on the nucleic DNA content and  ̄3H-TdR incorporation into DNA of PASMC cultured in NECCM. It is suggested that hypoxia may stimulate synthesis and secretion of some cytokinase which can stimulate the proliferation and the synthesis of collagen of PASMC and that RSM can inhibit this process.
