摘要
采用聚合酶链反应和寡核苷酸探针杂交技术,检测21例口腔粘膜鳞癌组织中Ha-ras癌基因第12位密码子G→T突变,结果有6例存在这种点突变(28.6%),而10例正常口腔粘膜组织无1例发生这种突变。提示:Ha-ras癌基因第12位密码子G→T突变可能导致口腔癌中癌基因的激活。并对点突变的临床意义进行了讨论。
The polymeras chain
reaction(PCR)was used to amplify the DNA sequence of Ha-ras oncogene
in the tissuses of oral mucosal squamous cell carcinoma and the G→T
mutation at codon 12 of the C-Ha-ras oncogene was detected by
hybridization with mutation specific oligonucleotide probes.The
results showed that the G→T mutation rate was 28.6%(6/21)In human
oral cancers,but no mutation was found in 10 normal oral mucosal
tissues.This study suggested that the G→T mutation at codon 12 of
the C-Ha-ras oncogene may lead to activation of C-Ha-ras oncogene.The
clinical significance of the mutation was also discussed.
