摘要
单核细胞趋化蛋白1(MCP-1)是一种强的单核细胞趋化因子,在体外可由多种细胞分泌,包括血管平滑肌细胞(SMC)。本研究采用斑点杂交方法检测了氧化修饰低密度脂蛋白(OX-LDL)、极低密度脂蛋白(VLDL)及氧化修饰极低密度脂蛋白(OX-VLDL)对培养的兔主动脉SMCMCP-1mRNA表达的作用。结果表明,培养的SMC可表达MCP-1mRNA,而且SMC暴露于OX-LDL、VLDL及OX-VLDL后,其MCP-1mRNA表达水平明显增高,分别增高约11倍、6倍和20倍。因此我们认为OX-LDL、VLDL及OX-VLDL可以刺激培养的兔主动脉SMCMCP-1mRNA的表达。
The migration of monocytes into subendothelial space is one of the earliest events in the pathogenesis of atherosclerosis.Monocyte chemotactic protein 1(MCP-1)is a potent monocyte chemo-attractant secreted by many cells in vitro,including vascular smooth muscle cells.In this study we have examined the effects of oxidatively modified LDL(OX-LDL),very low density lipoprotein(VLDL)and oxidativelv modified VLDL(OX-VLDL)on the expression of MCP-1 mRNA in cultured rabbit aortic smooth muscle cells(SMCs)using dot blotting analysis.The results showed that cultured SMCs could express MCP-1 mRNA and the levels of MCP-1 mRNA were increased significantly in SMCs when exposed to OX-LDL(11-fold),VLDL(6-fold)and OX-VLDL(20-fold).We conclude that OX-LDL,VLDL and OX-VLDL can stimulate the expression of MCP-1 mRNA in cultured rabbit aortic SMCs.
