摘要
报道了应用时间分辨免疫荧光氏分析血清中CEA的条件研究。用我们研制的N ̄1-(对-异硫氰基苄基)-二乙三胺-N ̄1,N ̄2,N ̄3,N ̄3-四乙酸铕为螯合剂,用Eu标记了抗CEA单克隆抗体(McAb)C_(17)。在免疫分析程序中,对包被的McAb的种类、浓度和封闭时间、第一和第二次反应的温育时间、标记C_(17)的浓度、分析缓冲液等主要实验条件,进行了实验最佳化选择。分析方法的灵敏度为1ng/ml。
The experimental conditions of detecting carcinoembryonic antigen (CEA)by means of time-resolved immunofluorometric assay are studied.A chelate-Eu ̄(3+)-N ̄1-(p-isothiocyanatobenzyl)-di-ethylenetriamine-N ̄1,N ̄2,N ̄3,N ̄3-tetraacetic acid synthesized is used to label mouse monoclonal antibody(McAb)C_(17)against CEA. In the assay,the CEA standard is first reacted with the coated McAb.Afterwashing ,the labeled C_(17) is added and the second reaction is followed.The important experimental con-ditions such as the coated McAbs a nd their concentration,block ing time ,incubation time for the firstand second reaction,the concentration of labeled C_(17) and the assay buffer solution,etc. are selectedand optimized in the assay procedures. the sensitivity of the assay is 1 ng/ml.
