鼠脑内注射rIL-2对脑组织和血脑屏障的影响

摘要 利用脑立体定向术向25只Wistar大白鼠右半球顶叶内注射重组白细胞介素-2(rIL-2),左半球相应的部位注射生理盐水作为对照,用辣根过氧化物酶(HRP)来确定血脑屏障损伤的范围,同时留取部分脑组织进行HE染色,观察12h,1d,3d,5d,8d的变化。结果表明:脑内注射rIL-2仅引起注射部位局部血脑屏障的改变,而单核细胞、巨噬细胞或粒细胞的滤出可能是维持技改变的一个因素,脑内局部注射rIL-2是安全的。 With the use of stereotaxic technique, the authors study the histological effects and BBB changes in twenty - five rats which occur after right parieal lobe intracerebral injection of rIL-2, the left same place with controls. Animals were killed at various intervals 12 hours, 1, 3, 5, 8 days, some brain tisses were sectioned and processed with HE stain for light microscopy. The authors can determine the injuries of BBB through marked neuronal cells and the region of HRP extravasation was regarded as the area of cerebral edema. The results suggest a single injection of rIL-2 into the brain of rats enhances cerebrovascular permeability in the vicinity of injection site which turns to help immure factors and cells into the brain and shows its efficacy. Mononuclear cells macrophages or granulocytes may cause persistent disruption of BBB. Injection of rIL-2into brain potentiates inflammatory cellular events occuring after cortical trauma, no direct toxicity occurs surounding needle track brain cell. It is suggested that intracerebral injection of rIL -2 is safe,and only the edema period is longer
