摘要
本文报告了以抗肾综合征出血热病毒(HFRSV)大鼠单克隆抗体(McAb)杂交瘤细胞系为模型,研究比较了用常规营养液添加培养法与静置细胞培养法(非添加培养)生产大鼠McAb的产量和滴度的效果。结果表明应用静置细胞培养法,McAb的产量明显高于添加细胞培养法2~3倍,抗体效价亦增加至少一个数量级,并且有简便易行、快速省时、减轻劳动强度和节省原材料的特点,值得试用。
Taking rat hybridoma cells secreted monoclonal antibody against hemorrhagic fever with renal syndrome virus (HFRSV) as the mold, we compared production and liter of McAbs in supernatant by two methods of hybridoma cell culture. The results showed concentration of McAbs by non re-fed cultures method was 2-3 times higher than by re-fed culture method. In addition ,the titer of mon-oclonal immunoglobulin (Ig) was once higher in thenon re-fed culture medium than in the renewed cul-ture. Besides, such non re-fed cultures also may savetime, decrease labour intension and reduce cost. So the results indicated the non re-fed culture was a simple and rapid method for in vitro increasing production of McAb. It can be used for hybridoma cell culture.
