摘要
以荧光探剂(DPH)标记细胞膜脂区,利用稳态荧光偏振技术观测硒对四氯化碳损伤的原代培养人胚肝细胞膜流动性的影响,同时检测了丙二醛及硒谷胱甘肽过氧化物酶(Se-GSH-Px)活性水平。结果表明,预加硒可有效地维护受损肝细胞膜流动性。其机制可能与增强Se-GSH-Px活性,抑制脂质过氧化有关,且此作用受预作用时间、剂量等因素的影响。
In order to study the effect of selenium on the of fluidity plasma membrane. human fetal hepatocytes in primary culture were pretreated with selenium and then exposed to carbon tetrachloride. The fluidity of hepatocytes plasma membrane was measured by the fluorescence polarization method using 1,6-dipheny-1,3,5-hexatriene as a fluorescentprobe. The results showed that the increase fluidity of and the activity of Se-GSH-Px, the decrease of MDA were related to the selenium concentration and protective time. It is suggested that selenium can enhance the activity of Se-GSH-Px, and inhibit the lipid peroxidation, and thus maintain the fluidity of human fetal hepatocytes plasma membrane.
