金鱼受精过程超微结构研究的快速半薄切片定位

摘要 介绍一种适于金鱼受精过程超微结构研究的快速半薄切片定位方法。经Epon812包理的受精卵,切0.15~0.5μm厚的切片,用甲苯胺兰O-派若宁B染液在酒精灯上烤染后镜检,整个过程在5min内完成。用此方法可将受精后至第一次卵裂前各时期的精子头、卵子染色体及雌雄原核找到并可进一步用于超微结构研究。 Embedded in Epon 812, the fertilized eggs of goldfish are cut into semithin sec-tions(0. 15~0.5 μm in thickness).Then,the sections are stained with toluidine blue O-pyronine B on spirit lamp for several seconds and examined under light mtcroscope. These proce-dures can be completed within five minutes. Using this method,we have successfully localizedthe sperm head,female chromosomes,and male and female pronuclei in eggs at yarious stagespostinsemination and preserved the samples for further ultrastructural studies.
