摘要
用光镜和电镜观察芦柑(Citrusreticulata)子叶在脱分化过程中的变化,结果表明,外植体脱分化由接触培养基切口处先启动,未培养的子叶薄壁细胞内充满脂类贮藏物,在脱分化过程中,贮藏物质逐渐减少直至消失,细胞核相对体积增加并常出现核膜内陷和核仁小泡,多聚核糖体与粗面内质网增加,线粒体在数量,形态、分布及嵴的发达程度上随着细胞的脱分化进程发生规律性变化。
changes during the course of dedifferentiation in cotyledon explants of citrusreticulata were investigated by light microscope and electron microscope. The results showed that dedif-ferentiation in exdants initially started at the cut in contact with media. Non-cuitured parenchymacells of cotyledons contained a lot of storage materials,lipids.During the course of dedifferentiation inexplants, storage materials gradually decreased and finally disappeared from the cells. The relativevolume of nuclei increased,and invagination of nuclear envelope and vesicles of nucleoli could be of-ten seen. Polyribosomes and rough endoplasmic reticulum increased. Mitochondria regularly changedin quantity,form,distribution and intemal structure with the development of dedifferlentiation of ex-plants.
