摘要
应用恒河猴做成亚慢性三硝基甲苯染毒(TNT)模型,在染毒前,染毒中期和试验表明咖啡因在动物体内的清除呈一级动力学,半衰期为8。88小时,这一结果说明咖啡因符合肝脏代谢模型药物的要求。TNT染毒后咖啡因清除率(CLca)下降,而同期血清甘胆醚(CA)浓度无变化。这一结果说明CLCa可能比CA能更早地反映接触TNT后的机体反应,染毒三个月后处死动物直接测定肝混合功能氧化酶(MFO)活性(P450及氨基比林脱甲基酶),发现咖啡因清除率和MFO有很好的相半性。由于MFO的变化往往是TNT接触后机体的早期的反映,因此这一结果为使用CLca作为TNT接触者的早期健康监护指标展示了一定的前景。
A subchronic intoxication model was made in 9 male rhesus monkey. The determination of caffeine clearance oate (Clca) was carried before giving Trinitroto(TNT),after givin TNT 1.5 and 3 months respectively. The result before intoxication showed that the clearance of caffeineacted presented and forstorder kinetics and half life time was 8.88 hrs, which indicated that cafeine that is mainly metabolinsed by liver corresponde with the demands of model compound for determinating the capacity of hepatic metabolition. CLca decreased after giving TNT but at same time no change of the glycocholic acid (GCA) in plasma, which indicated that CLca can reflected the earlier reaction of TNT toxicity. The determination of mixed functional oxidases (MFO) after giving TNT 3 monthes shiwed that CLca presented a good relationship to the MFOs. Since the change of MFOs in the earlier body reaction of the exposure to TNT,then CLca may act as the health monitoring index for the exposurers to TNT.
