摘要
采用Ames试验和大肠杆菌回变试验,对19种有机磷农药进行了诱变性研究,并分析了59种有机磷农药化学结构与诱变性的关系。结果19种受试有机磷农药在两种回变试验中获阳性结果的有5种,而且不论加与不加S9均不改变其定性结果。构效分析提示:三类构份的亚类改变可影响有机磷的诱变性,Y为链烃、X为O或S的甲基有机磷诱发回变的概率极高,X为O时诱变性最高。推测有机磷主要攻击鸟嘌呤的N-7位,通过甲基化作用引起碱基置换型突变。
Atotal of 19 organophosphorus pesticides(OPPs)were tested for mutagenicity by Ames test and Escherichia coli(E,coli)reversion assay,and the chemical structure and mutagenicity relationship of 59 OPPs were analyzed,Five out of the 19 OPPs give positive result in both reversion assay systems and the result were unchanged no matter the S9 is added in or not.The analysis of structure-activity relationship suggested that the mutagenicity ol OPPs could be affected by the subtype patterns changes of the three constituents,The incidence of reversion induced when Y as alkane and X as O or S was high,and it mainly attacked N-7 Position of quanine, and induced bass-pair substitution mutation through methylation.
