双温PCR法检测淋巴细胞白血病微小残留病 被引量:1

摘要 免疫球蛋白重链(IgH)重排可作为B淋巴系肿瘤的基因标志,应用PCR方法检测此重排可进行淋巴细胞白血病微小残留病(MRD)的研究。我们通过基因释放剂提取DNA及双温PCR循环,建立一种快速可靠PCR方法检测38例淋巴细胞白血病IgH重排基因,28/38(73.7%)阳性,敏感度为10 ̄(-4)~10 ̄(-5)水平。此方法简便、快捷且成本低,更适于临床检测实际需要。 Immunoglobulin heavy chain(IgH)gene rearrangement serves as a marker of clonality inB lymphoproliferative malignancies. PolymeraseChain reaction(PCR)can be used to detect this rear-rangement in the research of minimal residual disease(MRD).We report a simple and rapid PCR methodby using gene releaser to extract DNA and two-tem-perature PCR cycle to detect 38 ALL and CLL pa-tients. 28 patients(73.7%)were found with IgH re-arrangement,the sensitivity was between 10 ̄(-4) to10 ̄(-50).This method is rapid,simple,low cost and de-pendable,so it is more suitable for a routine servica-ble laboratory.
