摘要
本实验采用WEC技术(wholeembryoculture,全胚胎培养)探讨了As2O3不同时间对大鼠胚胎的致畸作用,染毒剂量为3.0μg/ml。结果表明:作用6h、16h对胚胎的生长发育和器官形态分化无明显影响;作用26h,反映胚胎生长发育和器官形态分化各指标伍均明显降低(P<0.05),41.7%的胚胎出现畸形;作用48h,改变更加明显,畸胎率达100%;畸形主要表现为前肢芽小或无、前后神经管未闭、无耳泡等。电镜观察可见VYS(脏层卵黄囊)上皮细胞顶端微绒毛变短、排列不规则和数量减少、线粒体肿胀、溶酶体数量减少等改变,提示VYS超级结构的改变可能与砷的致畸机制有关。
ammalian embryos were exposed at 3.0 μg/ml arsenic in
culture for 6,16,26 and 48hours by means of whole embryo culture technique.Six and 16 hours
exposure allowed em-bryos normal survival. However,the embryos exposed 26 and 48 hours
showed all parame-ters correlated with embryonic growth and morphological differentiation
were inhibited.Theincidence of abnormal embryos was 41.7%and 100%separately. The
abnormal embryoswere characterized by open neural tube,failure of development of limb buds
and sensory pla-codes. Under electron microscopy the embryonic visceral yolk
sac(VYS)showed the changesof ultrastructures of microvilli, mitochondria and lysosomes and
so on.These pathologicalchanges of VYS structures would be related to teratogenic mechanism
of arsenic.
