唾液富脯蛋白的分离提纯

Isolation and purification of proline-rich proteins from human parotid saliva

收集刺激性腮腺唾液１０００ｍｌ，利用先进的生化技术分离提纯出唾液富脯蛋白（ＰＲＰｓ）。经硫酸铵浓度差超速离心后得到初步纯化的ＰＲＰｓ，再经葡聚糖凝胶（Ｓｅｐｈａｄｅｘ）Ｇ－７５过滤和ＤＥＡＥＳｅｐｈａｄｅｘＡ－２５离子交换层析后可得到ＰＲＰｓ的６个组份，其分子量大小分别为１４３００、１０８００、１１０００、１２３５０、１３０００和１２５８０ｕ；等电点范围在４．１８～４．６０之间。６种组分的ＰＲＰｓ氨基酸组成分析，均富含脯氨酸、甘氨酸和谷氨酰胺。

In this study,we collected stimulatcd parotid saliva(1000ml),purified the PRPs from parotid saliva by some advanced biochemical technicalities.Crude PRPs could be obtained through(NH4)2SO4 ultra-centrifugation.Six purified PRPs were isolatcd after Gel filtration on a sephadex G-75 column and ion-change chromatography on a DEAE sephadex A-25 column.The molecular weight for PRPs were 14300,10800,11000,12350,13000 and 12580u respectively.The range of Amino acid composition showed that they all had a large amount of proline,glycine and glutamic acid.
