传代的人牙髓成纤维细胞在体外培养下的增殖和表型变化 被引量:1

摘要 观察和比较了在DMEM加10%FCS;DMEM加10%FCS和50μg/mlVitC和10mmol/Lβ-磷酸甘油钠两种培养条件下,第5代人牙髓细胞的某些生物学特征,观察时间为3、9、15、21d,通过ConA和WGA凝集素受体表达观察细胞的分化情况。结果表明第5代的人牙髓细胞可能是单一基因表型的细胞,其多层生长的能力减弱,牙髓细胞的凝集素受体(ConA和WGA)表达随培养时间延长未见明显改变。说明随着人牙髓细胞传代培养,人牙髓细胞的某些生物学特征发生了变化。 In order to study cellular and molecular biology of human dental pulp cells,pulp cells from human permanent molars were subcultured 5 times,the cells cultured with DMEM containing 10% FCS,or DMEM containing 10% FCS,plus 50μl/ml acorbic acid and 10mmol/ml sodium glycerophosphate,the cells were fixed for HE staining and immunohistochemical staining of WGA and ConA agglutinin receptor after 3,9,15,21d of incubation,the cells were examined by light microscopy and the following results were obtaines.The 5 times subcultured cells may be single-cell populations,the ability that the cells became multilayered were decreased,there were no difference in expression of ConA,WGA receptors of cells among different groups.It is therefore recommended that some cells were undergoing differentiation.
