HPLC DETERMINATION OF SULFADOXINE IN PLASMA CONTAINING PYRONARIDINE AND PRIMAQUINE

周效磺胺是临床上广泛使用的预防和治疗疟疾的药物,为增强抗疟效果常与咯萘啶、伯喹配伍应用。周效磺胺的血浓分析有分光光度法,微生物测定法,气相色谱法。

Sulfadoxine in plasma containing pyronaridine and primaquine was determined by HPLC. The acidified plasma was extracted with 5.0ml ethyl acetate. The organic extract was reduced to dryness, the residue was redissolved in mobile phase and an aliquit of this solution was then chromatographed on an ODS column. The mobile phase is CH_3OH-buffer solution (0.05mol/L citrus acid-Na_2HPO_4, pH=3) 40: 100, containing tri-n-butylamine 0.014mol/L and HClO_4 0.035mol/L. The detective limit of this method was 2ng, and the sensitivity was 0.2μg/ml. The linearity was demonstrated in the range of 1.0-10.0μg/ml. The average recovery was 87.68% and the CV of within-day and day-to-day was less than 5%.
