摘要
本文采用反相HPLC法测定阿霉素的血药浓度,血浆样品用甲醇-氯仿(1:4)在pH9.0的条件下提取后进样,5mmol/L磷酸-异丙醇-甲醇-乙腈(45:35:10:10,pH2.9)为流动相,荧光检测器λ_(ex)=450nm,λ_(em)=530mm。最低检测浓度10mg/ml,线性范围在30~1500ng/ml内r=0.9987,日内RSD为1.5%~2.4%,日间RSD为1.8%~3.7%。并对7例肺癌病例经支气管动脉灌注阿霉素的药代动力学进行了研究。
A RP-HPLC method for the determination of adriamycin in human plasma was reported.The ADM was extracted from 1ml of plasma at pH 9.0 by methanol-Trichloromethane(1:4).A solution of 5mmol/L orthophosphoric acid-propan-2-ol-methanol-acetonitrile(45:35:10:10)(pH2.9)was used as the mobile phase.The spectrofluorimeter for peaks detection with λ_(ex) =450nm,λ_(em)=530nm was used.The detection limit was 10ng/ml,the calibration curve was linear in the range from 30ng/ml to 1500ng/ml with r=0.9987.The RSD within a day were 1.5%~2.4%,and the RSD between days were 1.8%~3.7%.The method has been applied to study the pharmacokinetics of the adriamycin with intra-arterial infusion for 7 case of lung carcinoma.
