人脐带静脉内皮细胞的继代培养

A STUDY OF THE SUBCULTURE OF HUMAN ENDOTHELIAL CELL FROM UMBILICAL VEIN

人脐带静脉内皮细胞继代培养25代。应用光镜、电镜及Ⅷ因子相关抗原的免疫荧光方法进行鉴定与观察。在RPMI-1640培养液中加入20%人血清,内皮细胞生长因子200μg/ml,肝素100μg/ml及培养皿的明胶覆膜是人内皮细胞长期培养的重要条件。

Human endothelial cells derived from umbilical vein could be suboultured and survived 25 passages (double time 20-25 hours, 60-75 cumulative population doublings) . An observe was performed with light microscopy, electron microscopy and immunofluorescence microscopy using specific antisera aganist factor Ⅷ related antiggen. It identified that the cultured cells were endothelial cells Medium RPMI-1640 supplemented with 20% human serum, ondothelial cell growth factor 200μg/ml, heparin 100μg/ml and gelatin coated flasks were very important conditions for long-term culture of human endothelial cell.
