人胚气管器官培养及其在建立肺癌离体器官模型的应用 被引量:1

ORGAN CULTURE OF THE HUMAN FETAL TRACHEA AND ITS APPLICATION IN EXPERIMENTAL LUNG CARCINOMA

摘要 本实验在国内首次进行人胚气管器官培养,在此基础上以3、4苯胼芘作用,第一周时气管上皮细胞发生扁平鳞状化生,第二周出现上皮细胞复层增生和不典型增生等癌前病变。与利用鼠类气管培养进行致癌试验相比,人胚气管器官培养形成病变早而明显。人胚气管材料更适于建立实验性肺癌器官模型。 Tracheal rings of the normal 4 to 6 month fetus after artificial abortion within 8 hours were used to the organ culture and were treated with 6 μg benzo (a) pyrene per milliliter medium during 6 to 8 weeks of culture. Observation by light microscopy, and electronic transmission and scanning microscopy discovered that transform of the cultured treacheal rings of human fetus from the pseudostratified ciliated columnar epitholia into the simple squamous epithelia then into the stratified squamous epithelia was more rapid than that of hamster. Besides carcinogens Co-carcinogen should be used in older to produce a model for experimental lung carcinoma in the condition of the tracheal organ culture.
