摘要
本实验采用体外细胞培养法,直接观察了粉尘或药物对肺泡巨噬细胞的作用。发现:SiO_2尘粒使肺泡巨噬细胞的MDA产生明显增加,且为特异性;SOD和ALLO分别能抑制SiO_2所致的肺泡巨噬细胞的MDA产生的增加。
This article reported the effect of SiO_2 on the lipid peroxdation of alveolar macrophage in rat. The suspended rat alveolar maorophage were mixed with SiO_2 particles and TiO_2 particles respectively and incubated at 37℃ for 2 hrs, then the malonyldialdehyde(MDA) was determined. The results showed that MDA in SiO_2 group was significantly higher than that in normal control (normal saline, P<0.001), while the MDA in TiO_2 group showed no significant pifference compared with the normal control (P>0.05). The results suggested that the lipid superoxidate free radical reaction induced by SiO_2 might be determined by the molecular specificity of SiO_2. It was also found that, when free radical scavenger-superoxidate dismutase (SOD) and allopurinal (ALLO) were respectively added to the mixture of SiO_2 and suspended alvcolar macrophage, then incubated, SOD could significantly reduce the MDA production, so could the ALLO, but with a lesser degree. The results might indicate that SOD could prevent alveolar macrophage from the damage of lipid peroxidation induced by SiO_2, since ALLO is an inhibitor of xanthone oxidase, activated xanthone oxidase may also be one source of free radicals produced in silicosis.
