10株分泌HSV-Ⅱ型单克隆抗体的杂交瘤细胞系的建立

摘要 用HSV-ⅡUW268株免疫BALB/c鼠,取其脾细胞与小鼠骨髓瘤细胞融合,经筛选、克隆化后获得10株分泌抗HSV-ⅡUW268株单克隆抗体的细胞系。其中1株仅与HSV-ⅡUW268株发生特异免疫反应,9株与HSV-Ⅱ和HSV-IF_(17)均发生免疫反应。用免疫沉淀试验检测单克隆抗体的Ig类型为IgG_1。保存的杂交瘤细胞仍能稳定地分泌抗HSV-ⅡUW268株的单克隆抗体。 By immunizing BALB/c mouse with HSV-Ⅱ UW268 then hybridizing its spleen cell with myeloma cell, 10 strains of cell lines which could socret McAb were obtained (one strain could combine HSV-Ⅱ UW268 specially, nine others combined HSV-Ⅱ UW268 and HSV-Ⅰ F_(17)). Using immuno-preciptation the author determined that the Ig was of IgG_1 type.The preserved hybridoma could still secret McAb steadily.
