摘要
中间纤维、微管与微丝是胞质中的纤维状细胞器,对维持细胞外形、细胞器在细胞内的位置、调节细胞内与细胞间的运动等具有重要作用。角蛋白是直径为8~12nm的中间纤维。本文应用免疫荧光法研究了培养的正常人食管上皮,食管癌细胞与诱导分化前后食管癌细胞中间纤维角蛋白纤维的表达及其与张力原纤维的变化,以探讨角蛋白在肿瘤细胞分化与在肿瘤细胞生物学特性中的意义。
The expression of phenotypes of cultured cancer cells might be associated with cancer cell differentiation. By using the electron microscope and monoclonal antibodies against cytokeratins, PKK1, PKK2 and PKK3, the expression of cytokeratins and tonofilaments of normal embryonic esop-hageal epithelial cells and esophageal cancer cells were studied. Both PKK1 and PKK2 cytokeratins were detected in embryonic esophageal and esophag-eal cancer cells, but PKKs was not present. This indicated that these were squamous but not glandular epithelial cells. Also, a much greater degree of expression of PKK1 and PKK2, tonofilaments and desmosomes was observed in differentiated embryonic squamous esophageal epithelial cells and in DM-SO or retinamide induced differentiated esophageal cancer cells than in undifferentiated esophageal cancer cells.
