人麻风杆菌在裸鼠中的增殖 被引量:1

Multiplication of Mycobacterium Leprae in Nude Mice

摘要 本文报告了麻风杆菌在裸鼠(NIH/nu/nu)体内的大量增殖。麻风杆菌经足垫皮下接种(1.0×10~4)后,菌在裸鼠足垫中出现持续增高。至感染后21个月,足垫乎均菌量大于1.08×10~9,菌数增长了108000倍,每克组织含菌量大于5.4×10^(10);而平行接种的CFW系免疫正常小鼠的足垫平均菌量为3.68×10~?,表现为局限性增殖.麻风菌在裸鼠体内的生长对数期明显延长.对裸鼠体内增殖的抗酸杆菌(AFB)进行鉴定的结果表明;①不能在可培养分枝杆菌的培养基上生长;②吡啶提取试验失去抗酸性;③在鼠足垫试验中呈典型的麻风杆菌生长曲线,并对抗麻风药物如氨苯砜,利福平完全敏感。这些实验结果证明了在裸鼠体内增殖的AFB是人麻风杆菌. 从受感染的裸鼠组织病理学检查所见.被感染了的麻风杆菌也能播散到其他脏器和非接种部位;又从接种麻风菌的足垫部位的组织切片中见许多大小菌团;呈现瘤型麻风病变. 上述结果证明了裸鼠实验麻风模型的建立是成功的.对今后开展麻风病的防治研究工作更有促进作用. This article reported the multiplication of mycohacterium leprae in immune-deficient animals-nude mice (NIH/nu/nu). Mycobacterium leprae taken from multibacillary leprosy patient were inoculated with 1 .0× 104/foot pad into right hind foot pad(FP) of nude mice and CFW mice. The results of bacterial enumeration showed that acid-fast bacillus (AFB) in the nude mice continuously multiplied. At 21 months after inoculation, the numbers of AFB increased by over 108000-folds and reached>1 .08 × 108/FP (>5.4×1010/g tissues), while the CFW mice developed only 3.68×106/FP and exhibited their characteristic limited growth pattern. It is indicated that the phase of logarithmic growth was prolonged in nude mice. The AFB multiplied were identified as mycobacterium leprae by following identification test: (1) failure to growth on artificial media; (2) loss of acid-fastness by pyr-idine extraction; (3) typical growth curve of mycobacterium leprae in footpad and sensitivity to antileprosy drugs. The histopathological examination of nude mice foot-pad inoculated were found lepromatous lesions, in which enormous numbers of AFB were seen on 476 days postinoculation, AFB disseminated into the fear, nose, sciatic nerve, testicle, lymphonodes, liver, spleen, lung, and noninoculated foot-pad. As mentioned above, it is considered that experimental leprosy was established in nude mice.
