维生素A缺乏对大鼠红系祖细胞增殖分化的影响 被引量:25

摘要 采用造血祖细胞体外培养技术,较系统地观察维生素A缺乏(VAD)程度、病程及干预治疗对大鼠红系祖细胞增殖分化、外周血象、体重增长状况的影响。结果表明:1.早期红系祖细胞(BFU一E)对VAD最敏感,仅在边缘型或慢性VAD大鼠表现贫血;2.重度或急性VAD大鼠体重不增或下降出现较晚,外周血象在整个病程中一直正常,但BFU一E及晚期红系祖细胞(CFU一E)明显下降,BFU一E与血清VA成正相关;3.补充VA后,大鼠体重、外周血液学指标、BFU一E及CFU一E均明显增加。结果提示:VAD大鼠红系祖细胞增殖分化存在明显障碍,但这种损害可逆。 o study erythropoiesis associated with vitamin A(VA)deficient wea- ning rat,BFU-E CFU-E, hematological indicators and body weight gain were characterized.In experiment I,30 weaning rats were fed graded intak- es of VA for a 6-week comparison method.It was found that BFU-E was the most sensitive of the three parameters to VA deficiency and anemia was shown only in marginal or chronic VA deficiency.In experiment II,72 weaning rats were fed VA deficient diet for 10-weeks.The three para- meters described above were measured at the end of 2-week.4-week, 8-week and 10-week periods.Routine hematological indicators were normal,but BFU一Eand CFU一Ewere decreased and BFU一E was positively ralated to VA level in acute deficient rats.In experiment III,resupplement of VA to 45 deficient rats was tested.VA supplementation produced a direct positive response in body weight,routine hematological indicators,BFU-E and CFU- E。The above results suggested that the development of BFU-E and CFU- E in VA deficient rats were impaired but this impairment could be reversed。
